Research Article: Quercetin is a Potential Therapy for Rheumatoid Arthritis via Targeting Caspase- Through Ferroptosis and Pyroptosis
Abstract:
Rheumatoid arthritis is a chronic inflammatory autoimmune disease characterized by persistent synovitis and can destroy bone and cartilage tissue, leading to joint damage, chronic disability, and a shortened life span.– The global prevalence of RA is approximately %. The underlying molecular mechanism of its pathogenesis is still unknown, and the onset and progression of the disease are closely related to a variety of factors (genetic, environmental, and immunologic), among which an abnormal infiltration of immune cells has a significant impact on RA., Although significant progress has been made with several targeted immunotherapies, the limited therapeutic efficacy prompts us to investigate alternative pathogenic mechanisms and pathways, among which programmed cell death (PCD) is an innovative and major breakthrough. The imbalance of cell death in FLS and immune cell populations and the release of inflammatory cytokines ultimately promote synovitis, contributing to the RA’s deterioration. Apoptosis, autophagy, NETosis, necroptosis, ferroptosis, and pyroptosis are PCDs affecting the initiation and development of RA. RA may be an outcome of the interaction of multiple PCDs, and few studies have examined how ferroptosis and pyroptosis interact to exacerbate the course of RA. Thus, our study is looking for common disease targets.
