Abstract:
RADL is a member of the SWI/SNF chromatin remodeling protein family, which plays a crucial role in mobilizing nucleosomes and DNA-associated proteins within mammalian cells. Similar to other SWI/SNF remodeling enzymes, RADL possesses the ability to translocate along DNA, inducing superhelical torsion and promoting accessibility of nucleosomal DNA., The binding of RADL protein significantly enhances the stability of RAD nucleoprotein filaments formed on single-strand DNA (ssDNA) or double-strand DNA (dsDNA), thereby facilitating DNA strand exchange. Furthermore, RADL actively promotes the formation of specific dsDNA structures that effectively bind to RAD, facilitating the efficient repair and translocation of damaged DNA. Homologous recombination (HR), which involves the accurate repair of DNA double-strand breaks (DSBs) by utilizing intact sister chromatids as repair templates, relies heavily on the central role played by RADL.– Together with RAD, RADL constitutes essential components of the eukaryotic HR machinery. In vitro, RAD proteins assemble with ssDNA to form helical nucleoprotein filaments that facilitate the crucial step of DNA strand exchange during HR. In addition to its critical involvement in DNA damage repair (DDR), the absence of RADL leads to a significant increase in cell death resulting from DNA damage. Notably, RADL has been implicated in promoting the development and progression of various tumor types, including pancreatic cancer (PAAD), bladder cancer (BLCA), glioblastoma (GBM), and head and neck squamous cell carcinoma (HNSC), among others. Consequently, RADL emerges as a potentially important protein in tumorigenesis. However, existing studies on the role of RADL in tumors have primarily focused on specific cancer types, limiting our comprehensive understanding of its involvement in oncogenesis.
