Research Article: Exclusion of Ranitidine From Premedication Regimens During Paclitaxel Treatment

Abstract:
Paclitaxel is widely used as a chemotherapeutic agent. Its administration is complicated by hypersensitivity reactions (HSRs). These HSRs prompted the development of premedication regimens.1 Paclitaxel was subsequently authorized under the premise that premedication should be administered, consisting of several drugs, including an intravenous histamine-2 receptor antagonist (H2RA; eg, ranitidine). In 2019, contamination of ranitidine, a widely used H2RA, led to its market withdrawal.1,2 Promptly, clinicians ceased H2RA use in premedication regimens because of H2RA shortages. Currently, with sufficient formulations of H2RAs (eg, famotidine) available, clinicians are confronted with the dilemma of whether to adhere to the drug registration license or to keep H2RAs out of this regimen, as they have done in previous years.
