Research Article: Integrative Analyses of Pyrimidine Salvage Pathway-Related Genes Revealing the Associations Between UPP and Tumor Microenvironment
Abstract:
In non-malignant cells, nucleotide synthesis predominantly depends on the de novo synthesis pathway, which employs amino acids and glucose and is characterized by substantial ATP consumption. Interestingly, to circumvent the high energy requirements of replication stress, cancer cells can adapt by shifting their reliance from the de novo synthesis pathway mainly to the salvage pathways. Pyrimidine biosynthesis pathways have been viewed as promising targets, and a variety of agents have been developed and applied in cancer treatments., However, traditional agents that target nucleotide biosynthesis can result in undesirable outcomes due to the activation of nucleoside salvage pathways.
